以水稻hsp70基因cDNA为探针,从水稻基因组文库中筛选得到一个hsp70基因,并完成了该基因全长4794 bp的序列测定。该基因5′-端上游区含有一个5′-TATAAA-3′(TATA box)序列和一个高度保守的热激顺序元件5′-CTcgGAAcc-TICgAG-3′。由编码区核苷酸顺序推导出的水稻hsp70的氨基酸序列与其他植物hsp70的相比,有84%~92%的同源性。该基因在第72位氨基酸天冬氨酸的密码子处存在一个长为1939 bp的内含子,这是迄今为止发现的hsp70基因内含子中最长的。另外,在该基因3′-端非编码区内还存在一个57bp的内含子。Northern杂交结果显示该hsp70基因在正常温度(25℃)有表达,受热激处理后表达量增加。
A hsp70 gene was screened from the rice genomic library using rice hsp70 cDNA as a probe, and the full-length 4794 bp gene was sequenced. The 5’-terminal upstream region of the gene contains a 5’-TATAAA-3 ’(TATA box) sequence and a highly conserved heat-shock sequence element 5’-CTcgGAAcc-TICgAG-3’. The amino acid sequence of rice hsp70 deduced from the nucleotide sequence of the coding region showed 84% -92% homology with other plant hsp70. The gene contains a 1939 bp intron at the amino acid aspartic acid at position 72, the longest of the hsp70 introns found so far. In addition, a 57bp intron was also found in the 3’-terminal non-coding region of this gene. Northern blotting showed that the hsp70 gene was expressed at normal temperature (25 ℃) and increased after heat shock treatment.
