本文于体外进行了天然抗肿瘤药物三尖杉酯碱与牛血清白蛋白相互作用的研究,采用平衡透析法(pH=7.40±0.01,μ=0.2)在恒温25±1℃,振荡频率为110~120次/min,振荡16~18h的条件下,测定了其结合参数。实验结果表明,当三尖杉酯碱浓度为8.5×10~(-4)~1.55×10~(-3)mol/L,牛血清白蛋白浓度固定为4.5×10~(-4)mol/L时,其结合常数为3.51×10~2,结合部位数为0.93即三尖杉酯碱对牛血清白蛋白只有一个结合部位群。经荧光光谱法检测,证明该结合部位群在牛血清白蛋白分子的U部位。
In this paper, the interaction between natural antitumor drug harringtonine and bovine serum albumin was studied in vitro. The equilibrium dialysis method (pH=7.40±0.01, μ=0.2) was used at a constant temperature of 25±1°C and the oscillation frequency was 110. The binding parameters were measured under conditions of ~120 beats/min and shaking for 16 to 18 h. The experimental results show that when the concentration of harringtonine is 8.5×10 -4 -1.55×10 -3 mol/L, the concentration of bovine serum albumin is fixed at 4.5×10 -4 mol/L. At L, the binding constant was 3.51 x 10~2 and the number of binding sites was 0.93. That is, harringtonine had only one binding site group for bovine serum albumin. Detection by fluorescence spectroscopy proved that the binding site group is in the U site of the bovine serum albumin molecule.
