目的观察通心络胶囊对大鼠大脑中动脉栓塞后缺血半暗区生长相关蛋白-43(GAP-43)和突触素(Syp)表达的影响。方法将55只雄性 SD 大鼠随机分为通心络组、自然恢复组和假手术组。应用线栓法建立大鼠大脑中动脉闭塞模型(MCAO),以免疫组化及图像分析技术检测脑缺血后1、3、7、14、28 d 缺血半暗区 GAP-43和Syp 的表达。结果脑缺血后缺血半暗区 GAP-43表达自术后1、3 d 开始增高,第7 d达到高峰,自第14 d 开始降低。自然恢复组 Syp 于缺血后1 d 表达减少,3 d 达最低值,1周开始增高但不及正常表达数值,2、4周表达高于假手术对照组。通心络组缺血半暗区 GAP-43和 Syp 的表达分别在3 d 和1、2、4周时高于自然恢复组。结论通心络可促进卒中后缺血半暗区 GAP-43和 Syp 的表达,具有促进突触再建和增强、完善突触功能的作用。
Objective To observe the effect of Tongxinluo Capsule on the expression of growth-associated protein-43 (GAP-43) and synaptophysin (Syp) in ischemic penumbra after middle cerebral artery occlusion in rats. Methods Fifty-five male SD rats were randomly divided into Tongxinluo group, natural recovery group and sham operation group. A rat model of middle cerebral artery occlusion (MCAO) was established with a line occlusion method. Immunohistochemistry and image analysis techniques were used to detect GAP-43 and Syp in the ischemic penumbra region on 1, 3, 7, 14 and 28 days after cerebral ischemia. expression. Results After cerebral ischemia, the expression of GAP-43 in the ischemic penumbra began to increase at 1 and 3 days after operation, peaked at 7 days, and decreased from the 14th day. In the natural recovery group, Syp expression decreased at 1d after ischemia, reached the lowest at 3d, increased at 1st week but was not as normal, and was higher at 2 and 4 weeks than in the sham control group. The expression of GAP-43 and Syp in the ischemic penumbra group was higher than that in the natural recovery group at 3 days and 1, 2 and 4 weeks, respectively. Conclusion Tongxinluo can promote the expression of GAP-43 and Syp in post-stroke ischemic penumbra, and it can promote synapse reconstruction and enhance synaptic function.
