鼻咽癌(NPC)是中国南方常见的恶性肿瘤,NPC与EB病毒关系十分密切。通过免疫亲和层析的方法了解NPC肿瘤标本中p53与病毒蛋白或细胞蛋白间潜在的相互作用,这种相互作用可能引起p53在NPC组织中累积。建立单克隆抗体pAb1801,pAb240免疫亲和层析柱,从NPC转移淋巴结分离p53结合蛋白。银染和免疫印迹证实两种免疫亲和层析柱可高效纯化p53蛋白,并共纯化74kD和26kD蛋白。26kD蛋白可被抗p53抗体识别;74kD蛋白与p53蛋白以次级键结合,并与NPC患者血清产生微弱的阳性反应。
Nasopharyngeal carcinoma (NPC) is a common malignant tumor in southern China, and NPC is closely related to Epstein-Barr virus. The potential interaction between p53 and viral proteins or cellular proteins in NPC tumor samples is known by immunoaffinity chromatography and this interaction may cause the accumulation of p53 in NPC tissues. To establish monoclonal antibodies pAb1801, pAb240 immunoaffinity column, lymph node metastasis from NPC to separate p53-binding protein. Silver staining and immunoblotting confirmed that the two immunoaffinity columns could efficiently purify p53 protein and co-purify the 74kD and 26kD proteins. The 26kD protein is recognized by anti-p53 antibodies; the 74kD protein binds secondary to p53 protein and produces a weak positive reaction with NPC patient sera.
