This paper presents a reliable method for measuring red blood cell deformability in whole blood based on the Naclepore filtration method described by REID et al. (1976). The accuracy of the method is greatly improved due to the elimination of the air bubble technology trapped in the pores of the filter (the poor repeatability observed in the previous observation due to the bubbles trapped in the filter), so that the accuracy is within 5%. This paper presents a simple analysis of the characteristics of whole blood flow and hematocrit as a function of whole blood flow filtration time to eliminate the effects of different distributions of mid-hematocrit during leukocyte blockage and whole blood filtration. The average via time of a single red blood cell obtained can be used as a quantitative indicator of its deformation. In addition, the method takes full advantage of electronically controlled valves and suction pumps to allow repeatable measurements over a three-minute time interval. This improved filtration method is also suitable for clinical diagnosis, because it can avoid the experimenter’s blood sample
