采用检测K-ras基因第12位密码子点突变方式(CGT、GTT、GAT)设计的顺序特异性引物(SSP),对胰腺癌及慢性胰腺炎的冰冻组织和胰液进行PCR扩增,产物借助常规电泳及染色判断有无K-ras基因突变及突变方式。结果:胰腺癌组织及胰液K-ras基因突变率分别为95.1%(39/41)、94.1%(16/17),而慢性胰腺炎组织(29例)、胰液(17例)及正常胰腺组织(12例)、胰液(4例)均无K-ras基因突变.研究表明:PCR-SSP快速、简便、特异、敏感,可以作为术前鉴别慢性胰腺炎和胰腺癌的一种实用新方法。
Sequential specific primers (SSP) designed to detect point mutations (CGT, GTT, GAT) in the 12th codon of the K-ras gene were used to amplify the frozen tissue and pancreatic juice from pancreatic cancer and chronic pancreatitis. Routine electrophoresis and staining were used to determine whether K-ras gene mutations and mutations occurred. RESULTS: The mutation rates of K-ras gene in pancreatic cancer tissue and pancreatic juice were 95.1% (39/41) and 94.1% (16/17), respectively, while chronic pancreatitis (29 cases) and pancreatic juice (17 cases). There were no K-ras mutations in normal pancreatic tissue (12 cases) and pancreatic juice (4 cases). Studies have shown that PCR-SSP is rapid, simple, specific, and sensitive and can be used as a practical new method to identify chronic pancreatitis and pancreatic cancer before surgery.
