目的建立短暂性脑缺血发作(TIA)小鼠模型,探讨灯盏生脉胶囊(DZSM)对其神经保护作用及可能的机制。方法采用尾静脉注射t-BHP及低氧方法成功制备了TIA样发作小鼠模型,分别给予DZSM 0.36 g/kg、DZSM 0.18 g/kg、DZSM 0.09 g/kg灌胃治疗,进行神经功能评分及行为学检测,化学比色法检测脑组织SOD、MDA、GSH-px的含量。结果高、中、低剂量DZSM均可明显改善TIA小鼠神经功能缺损评分情况,DZSM 0.36 g/kg、DZSM 0.18 g/kg可明显改善TIA小鼠卒中指数。TIA小鼠血清MDA明显增高、SOD、GSH-Px含量明显下降,高、中、低剂量DZSM可使TIA小鼠脑MDA含量均明显下降,而GSH-Px、SOD含量明显上升。结论灯盏生脉胶囊可降低TIA小鼠的神经功能缺损评分和卒中指数,其作用机制可能通过提高TIA小鼠体内抗氧化系统的活性,降低脂质过氧化反应,从而抑制t-BHP过氧化反应导致的神经、血管内皮细胞等氧化应激损伤来实现。
Objective To establish a transient ischemic attack (TIA) mouse model to investigate the neuroprotective effect of DZSM and its possible mechanism. Methods The TIA-like seizure mouse model was successfully established by tail vein injection of t-BHP and hypoxia. DZSM 0.36 g / kg, DZSM 0.18 g / kg and DZSM 0.09 g / kg were given respectively. Behavioral testing and chemical colorimetry were used to detect the content of SOD, MDA and GSH-px in brain tissue. Results High, medium and low doses of DZSM can significantly improve the TIA mice neurological deficit score, DZSM 0.36 g / kg, DZSM 0.18 g / kg can significantly improve the TIA mouse stroke index. TIA mice serum MDA was significantly increased, SOD, GSH-Px content decreased significantly, high, medium and low doses of DZSM can make TIA mice brain MDA content decreased significantly, and GSH-Px, SOD increased significantly. Conclusion Dengzhan Shengmai Capsule can reduce the neurological deficit score and stroke index of TIA mice. The mechanism may be through the anti-oxidation system activity of TIA mice, reduce the lipid peroxidation, thereby inhibiting the t-BHP peroxidation Resulting in nerve, vascular endothelial cells and other oxidative stress to achieve.
