建立了大鼠血浆中胡椒碱浓度的UPLC测定法,并研究了不同给药介质对胡椒碱在大鼠体内的药动学特征的影响.大鼠灌胃给予不同给药介质的胡椒碱后,其药动学特征有明显区别,CMC-Na的浓度为0.3%时,胡椒碱达峰较慢,生物利用度较低;而CMC-Na的浓度为1%、3%时,胡椒碱的生物利用度较高,且相差不大,但Tmax和MRT相差较大,以植物油为给药介质灌胃给药后,胡椒碱在大鼠体内吸收缓慢,呈缓释效果,绝对生物利用度较低.因此在进行胡椒碱对其他药物增效作用的体内研究时,应选择适当的给药介质及给药时间.
The UPLC assay was established for the concentration of piperine in rat plasma and the effects of different administration media on the pharmacokinetics of piperine in rats were studied.After administration of piperine to different administration media, The pharmacokinetic characteristics were significantly different. When the concentration of CMC-Na was 0.3%, the peak of piperine was slow and the bioavailability was low. When the concentration of CMC-Na was 1% and 3%, piperine High utilization rate and little difference, but the difference between Tmax and MRT is larger. After the rats were given gavage with vegetable oil as medium, piperine absorbed slowly in rats with slow release effect and the absolute bioavailability was low Therefore, in the study of the synergistic effect of piperine on other drugs, we should choose the appropriate media and time of administration.
