目的:研究全反维甲酸(ATRA)等干扰因子对肺癌细胞株A549侵袭、增殖及ICAM-1分子表达的调节作用。方法:根据细胞穿过铺有Matrigel胶多孔膜数量检测细胞侵袭力,用改良龙胆紫吸收比色计数法检测细胞增殖,以流式细胞分析和酶联免疫吸附试验检测ICAM-1、sICAM-1表达水平。结果:A549 经ATRA处理后,细胞表面ICAM-1表达降低,体外侵袭力明显降低,体外增殖明显抑制;IFN α不增加细胞ICAM-1表达,但抑制体外增殖及体外侵袭力;IL-1 明显增强细胞表面ICAM-1表达及培养上清sICAM-1含量,体外侵袭力有增高趋势。结论:ATRA、IFN α、IL-1 对A549 细胞体外增殖及ICAM-1、sICAM-1表达有不同的调节作用,而对其转移潜能具有不同调变作用。
Objective: To study the regulation effect of all-anti-retinoic acid (ATRA) and other interfering factors on the invasion and proliferation of lung cancer cell line A549 and the expression of ICAM-1 molecule. Conclusion: ATRA, IFN α and IL-1 have different effects on the proliferation of A549 cells and the expression of ICAM-1 and sICAM-1 in vitro, but have different modulation effects on their metastatic potential.
