目的 探讨VEGF及其受体KDR在肝细胞肝癌 (HCC)的血管生成、生长及转移中的作用及p5 3、ras对VEGF表达的调控。方法 用免疫组织化学、原位杂交及图像分析的方法研究肝细胞肝癌 45例 ,肝硬化 2 1例及正常肝 8例的石蜡标本切片 ,从而探讨VEGF、KDR与肝癌病理学特点的关系及p5 3、ras对其表达的影响。结果 肝癌组中VEGF的表达明显高于肝硬化组和正常肝组 ,3组的表达率分别为 6 4% (2 9/4 5 )、14% (3/2 1)、12 % (1/8)。VEGF的表达与肝细胞肝癌的血管生成和转移显著相关 (r =0 46 0 7,P <0 0 5 ) ,p5 3可上调VEGF的表达 (P <0 0 5 ) ,而ras对其无影响 (P >0 0 5 )。KDR不仅表达于血管内皮细胞 ,还表达于肝癌细胞。结论 VEGF在肝细胞肝癌的血管生成、生长及转移中可能具有重要意义。
Objective To investigate the role of VEGF and its receptor KDR in angiogenesis, growth and metastasis of hepatocellular carcinoma (HCC) and the regulation of p53 and ras on VEGF expression. Methods Immunohistochemistry, in situ hybridization and image analysis were used to study 45 cases of hepatocellular carcinoma, 21 cases of liver cirrhosis, and 8 cases of normal liver. The relationship between VEGF, KDR and pathological features of hepatocellular carcinoma and p5 were studied. 3. The effect of ras on its expression. Results The expression of VEGF in the liver cancer group was significantly higher than that in the cirrhosis group and the normal liver group. The expression rates of the three groups were 6 4% (2 9/4 5 ), 14% (3/2 1), 12 % (1/ 8). The expression of VEGF was significantly correlated with the angiogenesis and metastasis of hepatocellular carcinoma (r = 0 46 0 7, P <0 05), p53 could up-regulate the expression of VEGF (P <0 05), but ras had no effect on it. (P > 0 0 5 ). KDR is not only expressed in vascular endothelial cells but also in hepatoma cells. Conclusion VEGF may play an important role in the angiogenesis, growth and metastasis of hepatocellular carcinoma.
