探讨转IFNγ基因膀胱癌瘤苗的可行性。采用脂质体介导法将携带鼠IFNγ基因的逆转录病毒载体转染入鼠膀胱癌细胞系BTT739中,免疫荧光法检测转基因细胞MHCI类抗原表达;动物实验观察其致瘤性和抗肿瘤免疫功能。结果显示:建立能自分泌IFNγ的膀胱癌瘤苗BTT739/IFNγ,瘤苗细胞MHCI类抗原表达量明显增加,致癌性显著降低,并可诱导机体产生特异性抗肿瘤免疫,CD+8T淋巴细胞在抗肿瘤免疫过程中起重要作用。结果证明:转IFNγ基因膀胱癌瘤苗可诱导机体产生特异性抗肿瘤免疫反应
To explore the feasibility of transfer of IFN γ gene bladder cancer vaccine. The retroviral vector carrying murine IFNγ gene was transfected into the mouse bladder cancer cell line BTT739 by liposomemediated method. The expression of MHC class I antigen in transgenic cells was detected by immunofluorescence. The tumorigenicity and anti-tumor activity Tumor immune function. The results showed that the expression of MHC class I antigen in tumor cells significantly increased after establishment of the bladder cancer vaccine BTT739 / IFNγ secreting IFNγ, the oncogenicity was significantly reduced, and the specific antitumor immunity was induced in the body.
