目的检测PTHLH在鼻咽癌细胞系及组织中的表达水平,从理论上初步探讨其在鼻咽癌发生发展过程中所起的作用。方法实时荧光定量PCR(FQ-PCR)检测PTHLH在7个鼻咽癌细胞系的表达情况,RT-PCR检测PTHLH在鼻咽癌及非癌鼻咽组织中的表达,免疫组化鉴定其在鼻咽癌及鼻咽上皮组织中蛋白水平的表达。结果PTHLH在鼻咽癌细胞系中的表达上调,以HONE1细胞中表达最高。RT-PCR分析显示PTHLH mRNA在鼻咽癌组织中表达明显上调(61.1%),免疫组化检测鼻咽癌石蜡标本(49例)发现,PTHLH蛋白在鼻咽癌组织中表达明显高于鼻咽上皮组织(12例)(P<0.001)。结论PTHLH基因在鼻咽癌细胞及组织中表达增高,提示其可能参与了鼻咽癌的发生发展,有望成为分子生物学诊断的靶点。
Objective To detect the expression of PTHLH in nasopharyngeal carcinoma cell lines and tissues and to explore its role in the development of nasopharyngeal carcinoma in theory. Methods The expression of PTHLH in seven nasopharyngeal carcinoma cell lines was detected by real-time fluorescence quantitative PCR (FQ-PCR). The expression of PTHLH in nasopharyngeal carcinoma and non-cancerous nasopharyngeal tissues was detected by RT-PCR. Pharyngeal carcinoma and nasopharyngeal epithelial protein expression. Results The expression of PTHLH was up-regulated in nasopharyngeal carcinoma cell lines and highest in HONE1 cells. RT-PCR analysis showed that the expression of PTHLH mRNA was significantly up-regulated in NPC tissues (61.1%). Immunohistochemical detection of NPC specimens (49 cases) showed that the expression of PTHLH protein in nasopharyngeal carcinoma tissues was significantly higher than that in nasopharyngeal tissues Epithelial tissue (12 cases) (P <0.001). Conclusion The increased expression of PTHLH gene in nasopharyngeal carcinoma cells and tissues suggests that it may be involved in the occurrence and development of nasopharyngeal carcinoma, which is expected to be the target of molecular biology diagnosis.
