以低温冰箱和液氮作为冷源,对四种淡水养殖鱼类的胚胎进行低温保存试验。采用10%二甲亚砜(DMSO)为抗冻剂,鲤鱼胚胎冷冻至-5℃、-10℃和-20℃,复温后的最高成活率分别为96%、90%和31%;团头鲂胚胎冷冻至-10℃、-15℃和-20℃,复温后的最高成活率分别为43%、20%和12.5%;青鱼胚胎冷冻至-5℃、-10℃和-11℃,复温后的最高成活率分别为100%、40%和0%;白鲢胚眙冷冻至-7℃,复温后的最高成活率为19.6%。造成胚胎损伤、致死的主要原因是胚胎在降温过程中细胞严重失水。
Cold storage experiments were conducted on embryos of four freshwater cultured fish using cold refrigerator and liquid nitrogen as cold sources. The carp embryos were frozen to -5 ℃, -10 ℃ and -20 ℃ with 10% dimethylsulfoxide (DMSO) as cryoprotectant. The highest survival rates after rewarming were 96%, 90% and 31%, respectively. The highest survival rates after rewarming were frozen to -10 ℃, -15 ℃ and -20 ℃ respectively, and the highest survival rates after rewarming were 43%, 20% and 12.5%, respectively. The green fishes embryos were frozen to -5 ℃, -10 ℃ and -11 ℃ , And the highest survival rates after rewarming were 100%, 40% and 0%, respectively; and the highest survival rate after rewarming was 19.6% after freezing to -7 ℃. The main cause of embryonic injury and lethal is the severe dehydration of the cells during the cooling process.
