目的:应用免疫组织化学方法检测不同时期血管瘤组织中胎盘生长因子(PLGF)及其受体(VEGFR-1)的表达,探讨其在血管瘤发生、发展过程中的作用机制。方法:采用免疫组织化学SP法检测50例皮肤血管瘤增生期、退化期及正常皮肤组织PLGF及VEGFR-1的表达水平,采用HPIAS-1000图文报告管理系统对PLGF和VEGFR-1的表达进行定量分析。结果:增生期组PLGF的表达明显高于退化期组和正常皮肤组(P<0.05),而后两组比较差异无统计学意义,P>0.05。增生期血管瘤血管内皮细胞中VEGFR-1呈高表达,正常皮肤组及退化组血管内皮细胞细胞中VEGFR-1弱表达。增生期组VEGFR-1的表达明显高于退化期组和正常皮肤组(P<0.05),而后两组比较差异无统计学意义,P>0.05。结论:PLGF和VEGFR-1在血管瘤增生期,可能发挥促进血管瘤内皮细胞过度增殖的作用,两者在血管瘤的发生、发展过程中起了重要的促进作用。
OBJECTIVE: To detect the expression of placental growth factor (PLGF) and its receptor (VEGFR-1) in different stages of hemangioma by immunohistochemistry, and to explore its mechanism in the occurrence and development of hemangiomas. Methods: Immunohistochemical SP method was used to detect the expression of PLGF and VEGFR-1 in 50 cases of proliferating, degenerative and normal skin tissues. The expression of PLGF and VEGFR-1 were detected by HPIAS-1000 graphic report management system Quantitative analysis. Results: The expression of PLGF in proliferative phase group was significantly higher than that in degenerative group and normal skin group (P <0.05), but there was no significant difference between the latter two groups (P> 0.05). VEGFR-1 was highly expressed in vascular endothelial cells of proliferative hemangiomas and VEGFR-1 was weakly expressed in vascular endothelial cells of normal skin and degenerative groups. The expression of VEGFR-1 in the proliferative phase group was significantly higher than that in the degenerative stage group and the normal skin group (P <0.05), but there was no significant difference between the two groups (P> 0.05). CONCLUSION: PLGF and VEGFR-1 may play an important role in promoting the proliferation of endothelial cells of hemangiomas during the proliferation of hemangiomas, both of which play an important role in the development and progression of hemangiomas.
