①目的 探讨五氯酚(PCP)对人类胎盘碱性磷酸酶(PLAP)的作用,为PCP中毒的防治提供理论依据。②方法 应用比色法分别测定有PCP和无PCP存在下PLAP的酶活力,根据其活力变化求出酶的相对活力;用双倒数作图法,确定PCP对PLAP的抑制类型并求其抑制常数(K_1)。③结果PCP的存在抑制了酶活力。当PCP的浓度在6mmol/L之内,随其浓度的增加而酶活力逐渐降低;当PCP的浓度为6mmol/L时,酶的相对活力为52.4%.PCP是PLAP的非竞争性抑制剂,其K_1为3.49mmol/L.④结论PCP对PLAP有抑制作。
Objective To investigate the effect of pentachlorophenol (PCP) on human placental alkaline phosphatase (PLAP) and provide a theoretical basis for the prevention and treatment of PCP poisoning. Methods The colorimetric method was used to determine the activity of PLAP in the presence of PCP and no PCP respectively. The relative activity of the enzyme was calculated according to the change of its vitality. The dual-reciprocal mapping method was used to determine the type of PLP inhibited by PCP and its inhibition constant (K_1). The results of the presence of PCP inhibited the enzyme activity. When the concentration of PCP was within 6mmol / L, the enzyme activity decreased with the increase of its concentration. When the concentration of PCP was 6mmol / L, the relative activity of enzyme was 52.4%. PCP is a noncompetitive inhibitor of PLAP, K_1 3.49mmol / L. Conclusion PCP inhibition of PLAP.
