目的观察冬凌草甲素(Ori)对小梁切除术后培养的兔结膜成纤维细胞(RCF)增殖的影响。设计实验研究。研究对象体外培养的小梁切除术后结膜滤过区来源RCF。方法制作兔眼小梁切除术模型,术后7天取兔滤过区结膜下组织进行成纤维细胞体外培养。分别用浓度依次为5、10、20、30、40、50μmol·L-1的Ori干预48小时。采用MTT法检测不同浓度药物对成纤维细胞生长的影响。主要指标成纤维细胞的光密度(OD)值和生长抑制率。结果加入Ori培养48小时后,5、10、20、30、40、50μmol·L-1不同浓度组成纤维细胞的OD值分别为(0.402±0.013)、(0.347±0.011)、(0.293±0.015)、(0.236±0.022)、(0.184±0.023)和(0.125±0.014),与无药对照组OD值(0.453±0.015)比较差异均有统计学意义(P=0.021~0.000),并呈浓度依赖性减小(F=5.48,P=0.003)。浓度为5、10、20、30、40、50μmol·L-1的Ori对成纤维细胞生长抑制率分别为11.26%、23.40%、35.32%、47.90%、59.38%和72.41%。结论Ori可以抑制培养的兔眼滤过区结膜成纤维细胞的增殖,抑制作用呈剂量依赖性。Ori可能成为一种治疗青光眼术后滤过泡纤维化的新药物。
Objective To observe the effect of Ori on the proliferation of rabbit conjunctival fibroblasts (RCF) cultured after trabeculectomy. Design experimental studies. The subject was in vitro cultured after trabeculectomy for conjunctival filtration zone-derived RCF. Methods The trabeculectomy model was established in rabbits. The subconjunctival tissues of the filtered area of rabbits were cultured in vitro on the 7th day after operation. The Ori was treated with concentrations of 5, 10, 20, 30, 40, and 50 μmol·L-1 for 48 hours. MTT assay was used to examine the effects of different concentrations of drugs on the growth of fibroblasts. The main indicators of fibroblasts optical density (OD) values and growth inhibition rate. Results After adding Ori culture for 48 hours, the OD values of fiber cells at different concentrations of 5, 10, 20, 30, 40, and 50 μmol·L-1 were (0.402±0.013), (0.347±0.011), and (0.293±0.015), respectively. , (0.236±0.022), (0.184±0.023), and (0.125±0.014) were significantly different from the OD value (0.453±0.015) in the non-drug control group (P=0.021-0.000), and were concentration dependent. The inhibition rates of Ori on fibroblasts at concentrations of 5, 10, 20, 30, 40, and 50 μmol·L-1 were 11.26%, 23.40%, 35.32%, 47.90%, 59.38%, and 72.41%, respectively. Conclusion Ori can inhibit the proliferation of cultured conjunctiva fibroblasts in cultured rabbit eyes in a dose-dependent manner. Ori may be a new drug for the treatment of fibrosis after filtering surgery for glaucoma.
