目的:应用响应面法优化离子液体微波辅助提取虎杖中芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚的提取工艺。方法:在单因素试验基础上,根据中心组合试验设计原理采用3因素5水平的响应面分析方法进行工艺优化,以5种蒽醌化合物总提取率为指标,进行综合评价。结果:优选出虎杖5种蒽醌类成分的最佳提取工艺为:加入40倍0.6 mol/L C8[MIM]Br,微波提取功率200 W,提取温度31℃,提取时间8 min;此工艺下5种蒽醌类成分总提取率为11.69 mg/g,理论值为11.74 mg/g。结论:该工艺提取时间短,操作简单,绿色环保,是快速高效提取虎杖蒽醌类物质的理想方法。
OBJECTIVE: To optimize the extraction process of aloe-emodin, rhein, emodin, chrysophanol and physcion in Polygonum cuspidatum by microwave-assisted extraction of ionic liquid with response surface methodology. Methods: Based on the single factor experiment, the process optimization was carried out by response surface analysis with 3 factors and 5 levels according to the design principle of central composite experiment. The total extraction rate of 5 anthraquinone compounds was used as an index for comprehensive evaluation. Results: The optimum extraction conditions of five anthraquinones from Polygonum cuspidatum were as follows: 40 mol 0.6 mol / L C8 [MIM] Br, microwave extraction power 200 W, extraction temperature 31 ℃ and extraction time 8 min. The total anthraquinone components extraction rate of 11.69 mg / g, the theoretical value of 11.74 mg / g. Conclusion: The process has the advantages of short extraction time, simple operation and environmental protection. It is an ideal method for rapid and efficient extraction of anthraquinones from Polygonum cuspidatum.
