目的 :研究非氰化物溶血剂在白细胞分类计数与血红蛋白测定中的作用 ,研制适用于 K450 0血液分析仪的配套试剂。方法 :用季铵盐型表面活性剂破坏红细胞 ,进行白细胞计数与分类 ,用 SLS破坏红细胞并进行血红蛋白的测定 ,以无机盐为主体组成中性等渗的稀释液 ,建立配套的 K450 0血液分析仪检测试剂。结果 :所形成的血红蛋白衍生物 λmax=535nm,且在 5h内稳定。自制试剂的精密度试验结果表明 ,各项主要参数的相对标准偏差 CV<5%。与 sysmex试剂可比性实验表明 ,两者结果无显著性差异。结论 :采用该试剂进行血常规分析 ,方法简便实用 ,结果准确 ,效果良好
Objective: To study the role of non-cyanide hemolytic agents in the differential leukocyte count and hemoglobin determination, and to develop suitable reagents for the K450 0 hematology analyzer. Methods: The erythrocytes were destroyed by quaternary ammonium salt type surfactant, and the white blood cell count and classification were performed. SLS was used to destroy the red blood cells and hemoglobin was measured. The inorganic salt was used as the main component to make a neutral isotonic diluent to establish the matching K450 0 blood analysis. Instrument detection reagents. Results: The hemoglobin derivative formed was λmax = 535 nm and was stable within 5 h. The precision test results of self-made reagents show that the relative standard deviation of each major parameter is CV<5%. Comparability experiments with sysmex reagents showed that there was no significant difference between the two results. Conclusion: This reagent is used for routine blood analysis. The method is simple and practical, with accurate results and good results.
