AIM: To investigate the protective effect of GW4064 on fructose-induced non-alcoholic fatty liver disease (NAFLD) in mice. Methods: The mice were randomly divided into normal control group, model group and treatment group. The rats in the model group and the treatment group were treated with 30% fructose water. After 8 weeks, the mice in the treatment group were orally administered with GW4064 (30 mg · kg -1) The levels of serum glucose, insulin, low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C), triglyceride (TG) and cholesterol (CHO) . Results: Compared with NAFLD model group, the levels of fasting blood glucose and insulin in GW4064 mice decreased significantly. The levels of TG, CHO and LDL-C in serum were significantly decreased and the levels of HDL-C were significantly increased. Pathological results show that GW4064 can significantly reduce hepatic steatosis in NAFLD mice and significantly reduce the degree of liver damage. CONCLUSION: NAFLD can be induced successfully with 30% fructose and GW4064 improves NAFLD by decreasing lipid deposition and increasing insulin sensitivity.
