目的探讨血管紧张素Ⅱ(AngⅡ)和白介素6(IL-6)在静脉留置针致血管损伤中的作用。方法选新西兰大耳兔40只,随机分为4组,每组10只,其中一组作为对照组,常规饲养1 W后留取静脉血和双耳外缘中段静脉血管组织。其余3组为实验组,兔双耳外缘静脉留置针穿刺置管,注射生理盐水,20 mL/次,1次/d。3组分别留置3 d、5 d、7 d,达到留置时间后抽取静脉血,麻醉后留取局部血管组织。用放射免疫法检测血液及血管组织中的AngⅡ和IL-6的含量。结果血液检测:AngⅡ含量,5d组、7d组明显高于对照组(P<0.01);实验组组间比较差异有统计学意义(P<0.01,P<0.05),随着留置时间的延长AngⅡ依次增高;IL-6含量,7 d组明显高于对照组、3 d组和5 d组(P<0.01)。组织检测:AngⅡ含量,3组实验组与对照组比较差异有统计学意义(P<0.01),实验组组间比较,5 d组、7 d组高于3 d组(P<0.05);IL-6含量,5 d组、7 d组明显高于对照组(P<0.01);实验组组间比较差异有统计学意义(P<0.01)。结论静脉留置针刺激可使AngⅡ和IL-6含量增高,引起血管收缩痉挛,促使血管炎症反应发生,而致血管损伤。
Objective To investigate the role of angiotensin Ⅱ (AngⅡ) and interleukin-6 (IL-6) in vascular injury induced by venous indwelling needle. Methods Forty New Zealand big-eared rabbits were randomly divided into 4 groups with 10 rats in each group. One group served as the control group, and venous blood and middle peripheral venous blood vessels of both ears were collected after routine feeding for 1 w. The other three groups were experimental group, rabbits in both limbs of the outer edge of the vein indwelling needle puncture catheter, saline injection, 20 mL / time, 1 time / d. 3 groups were left for 3 d, 5 d, 7 d, after the retention time to extract venous blood, local vascular tissue after anesthesia. The levels of AngⅡ and IL-6 in blood and blood vessels were detected by radioimmunoassay. Results Blood levels of AngⅡ were significantly higher in the 5d and 7d groups than those in the control group (P <0.01). There was significant difference between the two groups (P <0.01, P <0.05). With the prolongation of retention time, (P <0.01). The level of IL-6 in 7 d group was significantly higher than that in control group, 3 d group and 5 d group (P <0.01). The level of AngⅡ in the experimental group was significantly higher than that in the control group (P <0.01). The levels of AngⅡ in the experimental group were significantly higher than those in the control group at 5 d and 7 d (P <0.05) -6, 5 d and 7 d groups were significantly higher than those of the control group (P <0.01). There was significant difference between the experimental groups (P <0.01). Conclusion Intravenous infusion of acupuncture can increase the content of Ang Ⅱ and IL-6, causing vasoconstriction spasm, promoting blood vessel inflammation and vascular injury.
