目的:探讨一氧化氮(NO)和氧自由基(O-2)对培养大鼠皮层神经细胞的损伤作用及两种损伤因素间的关系。方法:应用原代大鼠皮层神经细胞培养方法,研究外源性损伤因素的作用。结果:①以硝普钠(SNP)为NO供体的损伤组较对照组的乳酸脱氢酶(LDH)释放量显著增高,而细胞存活率显著降低,加入SOD后SNP的损伤作用减轻。②O-2损伤组的结果与NO损伤组结果相似,同时应用O-2和NO组对神经细胞的损伤作用显著高于单独应用O-2组或NO组。结论:NO和O-2都能造成培养的大鼠皮层神经细胞损伤,并且二者之间存在协同作用
Objective: To investigate the effect of nitric oxide (NO) and oxygen free radicals (O-2) on the injury of cultured rat cortical neurons and the relationship between the two injury factors. Methods: Primary cultured rat cortical neuronal cultures were used to study the effects of exogenous injury factors. Results: ① Compared with the control group, the SNP-NO donor group had significantly higher lactate dehydrogenase (LDH) release, while the cell survival rate was significantly lower. The injury of SNP was alleviated after adding SOD. ② The results of O-2 injury group were similar to those of NO injury group. The damage of nerve cells by O-2 and NO groups was significantly higher than that of O-2 or NO alone group. CONCLUSION: Both NO and O-2 can cause injury of cultured rat cortical neurons and there is a synergistic effect between them
