植物特有的NAC转录因子在植物的生长发育、胁迫应答和激素调节等方面具有重要功能。本研究从橡胶树胶乳cDNA中克隆了HbNAC33基因,生物信息学分析显示该基因的完整开放阅读框(ORF)为1 092 bp,编码363个氨基酸。HbNAC33蛋白的N-端第3~156位氨基酸之间含有一个典型的NAC结构域,酵母试验表明,HbNAC33具有转录激活活性,转录激活区在C-端,具备了NAC转录因子的基本特征。序列比对和系统进化分析表明HbNAC33蛋白属于NAC转录因子家族中的ANAC011亚族。实时荧光定量PCR分析表明,HbNAC33在叶中的表达量显著高于其他部位;割胶,乙烯利(ET)和茉莉酸(JA)处理均能诱导HbNAC33的表达上调。以上结果表明,HbNAC33可能参与植物的生长发育和胁迫应答过程。
Plant-specific NAC transcription factors play an important role in plant growth and development, stress response and hormone regulation. In this study, HbNAC33 gene was cloned from rubber latex cDNA. Bioinformatics analysis showed that the ORF of this gene was 1 092 bp, encoding 363 amino acids. HbNAC33 contains a typical NAC domain between amino acids 3 ~ 156 of N-terminal. The yeast test showed that HbNAC33 has transcriptional activation activity and transcriptional activation region at the C-terminal, which possesses the basic features of NAC transcription factor. Sequence alignment and phylogenetic analysis indicated that the HbNAC33 protein belongs to the ANAC011 subfamily of NAC transcription factor families. Real-time PCR analysis showed that the expression of HbNAC33 in leaves was significantly higher than that in other parts of the leaves. HbNAC33 expression was up-regulated by tapping, ethephon (ET) and jasmonic acid (JA) treatment. The above results show that HbNAC33 may be involved in plant growth and stress response process.
