目的:研究儿茶素(catechin,Cat)对重组大鼠白细胞介素1α(rIL-1α)诱导佐剂性关节炎(AA)大鼠成纤维样滑膜细胞(FLS)功能的影响,并探讨其部分机制。方法:采用弗氏完全佐剂(FCA)诱导大鼠AA模型,分离培养大鼠成纤维样滑膜细胞,用MTT法检测FLS的增殖能力,放射免疫法(R IA)测定FLS产生PGE2和cAMP水平。结果:选择10 ng.mL-1为rIL-1α的最佳刺激浓度,Cat(100,50,25 mg.mL-1)体外给药可不同程度地抑制rIL-1α对AA大鼠FLS的促增殖反应,明显下调升高的PGE2的水平,明显上调降低的cAMP水平。结论:rIL-1α可以促进AA大鼠FLS的增殖,降低cAMP,升高PGE2,Cat能明显逆转rIL-1α对FLS的影响。
AIM: To investigate the effect of catechin (Cat) on fibroblast-like synoviocytes (FLS) function in rats with adjuvant arthritis (AA) induced by recombinant rat interleukin-1α (rIL-1α) Some of its mechanisms. Methods: Rat AA model was induced by complete Freund ’s adjuvant (FCA). Fibroblast - like synoviocytes of rats were isolated and cultured. The proliferation of FLS was detected by MTT assay. The levels of PGE2 and cAMP were determined by radioimmunoassay (RIA) Level. Results: Cathepsin (100, 50, 25 mg.mL-1) in vitro could inhibit the activation of FLS in rL-1α to some extent Proliferation response, significantly decreased the level of elevated PGE2, and significantly increased the level of decreased cAMP. Conclusion: rIL-1α can promote FLS proliferation, decrease cAMP and increase PGE2 in AA rats. Cat can significantly reverse the effect of rIL-1α on FLS.
