目的 研究 17β 雌二醇对缺血性脑损害的神经保护作用是否与Bc1 2蛋白的表达有关。 方法 采用大鼠大脑中动脉闭塞制成局灶性脑缺血模型。在缺血 2小时 ,再灌注 2 2小时后立即断头取脑 ,应用TTC染色和免疫组化方法 ,观察脑梗死体积及Bc1 2蛋白的表达。结果 17β 雌二醇用药组较对照组脑梗死体积显著缩小 (P <0 .0 5 ) ;Bc1 2蛋白的表达较对照组明显增强 (P <0 .0 5 )。结论 17β 雌二醇上调大鼠脑缺血时bc1 2的表达很可能是其具有神经保护作用的机制之一。
Objective To investigate whether the neuroprotective effect of 17β-estradiol on ischemic brain damage is related to the expression of Bcl-2 protein. Methods The middle cerebral artery occlusion rats were used to make focal cerebral ischemia model. The brain was decapitated at 2 hours after ischemia and 2 hours after reperfusion. The volume of cerebral infarction and the expression of Bcl-2 protein were observed by TTC staining and immunohistochemistry. Results Compared with the control group, the volume of cerebral infarction in 17β-estradiol treatment group was significantly reduced (P <0.05); the expression of Bc1 2 protein was significantly increased (P <0.05) in the control group. Conclusion The up-regulation of bc1 2 expression by 17β-estradiol in rats with cerebral ischemia may be one of the mechanisms of its neuroprotective effect.
