为提高生物素-亲和素系统对检测抗原或抗体的灵敏度和特异性,本文参照国外文献和自己设计的工艺,以活化生物素为原料,通过二步反应制得活化长臂生物素。其化学特性基本符合文献报道。用其标记羊抗兔Ig并与常用的活化生物素(短臂)作对比,结果表明在ABC-ELISA中,使用长臂生物素作标记物,可使特异性显色明显增强,标记的抗体稀释度可提高4~8倍,有助于降低BAS试剂本身的非特异性,提高检测的特异性和敏感性。
In order to improve the sensitivity and specificity of biotin-avidin system for detection of antigens or antibodies, this paper refers to foreign literature and their own design process to activate biotin by two-step activation of activated biotin. Its chemical properties basically in line with the literature. The sheep anti-rabbit Ig was labeled and compared with the commonly used activated biotin (short arm). The results showed that in the ABC-ELISA, the long-arm biotin was used as a marker to significantly enhance the specific color. The labeled antibody Dilution can be increased by 4 to 8 times, helping to reduce the non-specificity of BAS reagent itself and improve the specificity and sensitivity of detection.
