目的:初步研究麦角甾醇(ergosterol,ES)对肝癌细胞HepG2增殖的影响及可能机制。方法:实验设立对照组和麦角甾醇10μg/ml、50μg/ml、100μg/ml、200μg/ml组,用MTT方法和CSFE染色流式细胞术观察麦角甾醇对HepG2细胞增殖的影响,RT-qPCR检测p21mRNA表达,Western blot检测p21蛋白的表达影响。结果:与对照组比较,作用48h、72h后,麦角甾醇50μg/ml、100μg/ml、200μg/ml均对HepG2有明显的抑制增殖作用,p21 mRNA和蛋白的表达量均明显增高。结论:麦角甾醇在50~200μg/ml可以抑制HepG2细胞增殖,其机制可能与增高p21mRNA和蛋白表达相关。
Objective: To investigate the effects of ergosterol (ES) on the proliferation of HepG2 hepatocellular carcinoma cells and its possible mechanism. Methods: The effects of ergosterol on the proliferation of HepG2 cells were observed by MTT assay and CSFE staining. The mRNA and protein levels of ergosterol in HepG2 cells were detected by RT-qPCR p21mRNA expression, Western blot detection p21 protein expression. Results: Compared with the control group, ergosterol 50μg / ml, 100μg / ml and 200μg / ml all inhibited the proliferation of HepG2 at 48h and 72h, and the expression of p21 mRNA and protein were significantly increased. Conclusion: Ergosterol can inhibit the proliferation of HepG2 cells at 50 ~ 200μg / ml, which may be related to the increase of p21 mRNA and protein expression.
