目的研究天然化合物青藤碱对人前列腺癌DU145细胞增殖及凋亡的影响。方法采用MTT法检测青藤碱对DU145细胞增殖的影响;采用流式细胞仪AnnexinV/PI双染法检测青藤碱对DU145细胞凋亡的影响;采用电子显微镜观察青藤碱作用DU145细胞后细胞超微结构的变化。结果 MTT结果显示青藤碱浓度在100~400μmol/L时,作用24、48小时后,细胞增殖受到抑制,呈现明显时间剂量效应关系;流式细胞仪AnnexinV/PI双染法显示青藤碱(0、200、400μmol/L)作用于细胞24小时后,细胞凋亡率升高,且呈剂量效应关系;电子显微镜下青藤碱处理组细胞可见染色质浓聚、边集等典型的凋亡改变,而空白对照组没有相应的变化。结论青藤碱能有效抑制人前列腺癌DU145细胞增殖、诱导细胞凋亡。
Objective To study the effect of sinomenine, a natural compound, on the proliferation and apoptosis of human prostate cancer DU145 cells.
