[目的 ]用 PCR法检测嗜人 T细胞白血病病毒 I型 (HTL V- I)的分离情况。 [方法 ]采集 2例日本鹿儿岛县成人 T淋巴细胞白血病 (AL T)病人的肝素抗凝血 ,分离外周血单核细胞 (PBMCs) ,加入植物血凝素和白介素 - 2 ,与健康人 PBMCs共培养进行 HTL V- I的分离 ,然后抽提 DNA,用 HTL V- I的 gag1和 tax1两引物进行 PCR扩增 ,对扩增产物进行琼脂糖凝胶电泳确定病原分离结果。 [结果 ]2株都扩增出了 HTL V- I前病毒 DNA的 gag1和 tax1片断 ,证明了嗜人 T细胞白血病病毒 I型的分离成功。 [结论 ]PCR法是快速、准确检测嗜人 T细胞白血病病毒感染的有效方法之一。
[Objective] To detect the separation of human T cell leukemia virus type I (HTL V-I) by PCR. [Method] The heparin anticoagulant blood was collected from 2 patients with adult T - cell lymphocytic leukemia (AL T) in Kagoshima prefecture, Japan. Peripheral blood mononuclear cells (PBMCs) were isolated from peripheral blood mononuclear cells (PBMCs), and then added with phytohemagglutinin and interleukin - PBMCs were co-cultured to isolate HTL V-I. DNA was then extracted and PCR amplified with two primers, gag1 and tax1, of HTL V-I. The amplified products were subjected to agarose gel electrophoresis to confirm the isolation results. [Result] The gag1 and tax1 fragments of HTL V-I provirus DNA were amplified in both strains, which proved the successful isolation of type I human T-cell leukemia virus. [Conclusion] PCR method is one of the effective methods for rapid and accurate detection of human T-cell leukemia virus infection.
