目的 探讨丙戊酸钠 (VAP)对戊四氮 (PTZ)致疒间 大鼠海马Bax和Bcl 2表达的影响。方法 将2 4只成年Wistar大鼠随机分为正常对照 (NC)组、PTZ组和VAP组 ,PTZ组和VAP组大鼠腹腔注射阈下剂量的PTZ 35mg/ (kg·d) ,直至达到点燃标准。点燃后 ,VAP组大鼠经腹腔注入VAP 15mg/ (kg·d) ,PTZ组大鼠经腹腔注入生理盐水 ,30min后 ,再腹腔注射PTZ诱发癫疒间 发作。应用免疫组化法检测大鼠海马神经元Bax和Bcl 2蛋白的表达。结果 大鼠海马神经元Bax阳性细胞数和光密度 ,PTZ组均明显高于VAP组和NC组 (均P <0 0 1) ,VAP组明显高于NC组 (P <0 0 5 ) ;大鼠海马神经元Bcl 2阳性细胞数和光密度 ,PTZ组明显高于NC组(P <0 0 5 ) ,VAP组均明显高于PTZ组和NC组 (均P <0 0 1)。结论 VAP可以增强癫疒间大鼠海马神经元Bcl 2的表达和降低Bax的表达 ,有对抗癫疒间 发作导致细胞凋亡的作用。
Objective To investigate the effect of sodium valproate (VPA) on the expression of Bax and Bcl 2 in the hippocampus of PTZ induced rats. Methods Twenty-four adult Wistar rats were randomly divided into normal control group (NC), PTZ group and VAP group. PTZ group and VAP group were injected intraperitoneally with subliminal dose of PTZ 35 mg / (kg · d) until they were ignited standard. After ignition, rats in VAP group were injected intraperitoneally with VAP 15 mg / (kg · d). Rats in PTZ group were injected with normal saline through the abdominal cavity. After 30 minutes, intraperitoneal injection of PTZ induced seizures. Immunohistochemistry was used to detect the expression of Bax and Bcl 2 protein in hippocampal neurons of rats. Results The number of Bax positive cells and optical density in hippocampal neurons of rats in PTZ group were significantly higher than those in VAP group and NC group (all P <0.01), significantly higher in VAP group than in NC group (P <0.05) The number of Bcl 2 -positive cells and optical density in hippocampal neurons were significantly higher in PTZ group than in NC group (P <0.05), and were significantly higher in VAP group than those in PTZ group and NC group (all P <0.01). Conclusion VAP can enhance the expression of Bcl-2 and decrease the expression of Bax in the hippocampal neurons of the epileptic rats, and has the effect of antagonizing the epileptic seizure-induced apoptosis.
