目的探讨E-钙粘蛋白(E-cadherin)和基质金属蛋白酶2(MMP-2)的蛋白表达与大肠癌的关系。方法应用免疫组织化学技术检测E-cadherin和MMP-2蛋白在14例大肠腺瘤组织、102例大肠癌组织及相应癌旁正常粘膜的表达情况,分析二者表达与大肠癌发生发展的关系以及二者在大肠癌表达的相关性。结果 E-cadherin在大肠癌旁的正常组织中阳性率为68.6%,在癌组织中阳性率为42.2%,表达明显降低(P<0.01);相反MMP-2蛋白在癌组织中阳性率为50.0%,在正常组织中阳性率为13.7%,表达明显增高(P<0.01);Spearman等级相关分析结果显示两者在大肠癌组织中表达呈现负相关(rs=-0.269,P<0.01)。结论 E-cadherin的表达降低和MMP-2的表达增高与大肠癌的生物学行为密切相关,两者表达水平的高低可作为临床判断大肠癌预后有价值的指标。
Objective To investigate the relationship between the expression of E-cadherin and matrix metalloproteinase-2 (MMP-2) and colorectal cancer. Methods Immunohistochemistry was used to detect the expression of E-cadherin and MMP-2 protein in 14 cases of colorectal adenoma, 102 cases of colorectal carcinoma and their adjacent normal mucosa. The relationship between the expression of E-cadherin and MMP-2 protein and the occurrence and development of colorectal cancer were analyzed. The correlation between them in colorectal cancer. Results The positive rate of E-cadherin was 68.6% in normal tissues adjacent to colorectal cancer and 42.2% in cancerous tissues (P <0.01). The positive rate of MMP-2 protein in cancerous tissues was 50.0 %. The positive rate was 13.7% in normal tissues and the expression was significantly increased (P <0.01). Spearman rank correlation analysis showed that there was a negative correlation between the two expressions in colorectal cancer (rs = -0.269, P <0.01). Conclusions The decreased expression of E-cadherin and the increased expression of MMP-2 are closely related to the biological behavior of colorectal cancer. The expression levels of E-cadherin and MMP-2 may be used as a valuable index to evaluate the prognosis of colorectal cancer.
