目的:研究ADA蛋白质对ALKA基因表达的调节,探讨ADA蛋白质与ALKA基因有效转录的关系.方法:采用DNA顺序测定和体外转录的方法,以LAC基因启动子为对照进行实验.结果:ADA蛋白质加入以突变和野生ALKA基因启动子为模板的体外转录体系中,产生的ALKAMRNA高于无ADA蛋白质组;ADA蛋白质加入LAC基因启动子转录体系中,产生的LACMRNA与未加ADA蛋白质组基本相同.结论:ADA蛋白质对LAC基因启动子转录调节作用不明显,对于突变的和野生的ALKA基因转录均有促进作用,这种调节作用可能发生在启动子上游区.“,”Objective: We investigated the relationship between Ada protein and the effective transcription of alkA gene in order to study the regulation of alkA gene expression by Ada protein.
