基于一维微流控微珠阵列芯片,通过在微珠表面进行核酸功能化修饰,发展了一种新型的肿瘤转移相关基因检测平台.该芯片灵敏度高(DNA检测下限为0.02nmol/L),无需扩增即可直接对多种基因mRNA进行同时检测.以来源于同一病人大肠腺癌的原发癌和转移癌细胞为研究对象,成功地对两种癌细胞中多个肿瘤转移相关基因转录水平的表达进行了检测,并用RT-PCR验证了该芯片的检测结果.结果表明,该芯片具有样品及试剂消耗极小、灵敏度高、分析速度快、成本低等优点,并且可以实现高通量分析,在肿瘤转移早期诊断及机理研究等方面具有重要的应用价值.
