β-catenin基因是生物进化上高度保守的基因,是Wnt/β-catenin信号通路中的重要组分。本研究通过RACE方法获得了棉铃虫β-catenin基因的全长c DNA序列,命名为Har-β-catenin(Gen Bank登录号为KJ206237)。其开放阅读框长2382 bp,编码793个氨基酸残基。与已报道的其它物种β-catenin蛋白相似,Har-β-catenin蛋白具有多个ARM重复结构域。运用RT-PCR的方法比较滞育和非滞育蛹脑中Har-β-catenin基因的表达情况,结果表明非滞育蛹脑中Har-β-catenin基因的表达水平高于滞育蛹脑。最后,我们构建了包含Har-β-catenin全长编码区的真核表达载体并调查了Har-β-catenin的亚细胞定位情况。这些结论为我们进一步研究Wnt/β-catenin信号通路在棉铃虫滞育中的作用奠定了基础。
The β-catenin gene is a gene that is highly conserved in biology and is an important component of the Wnt / β-catenin signaling pathway. In this study, the full-length c DNA sequence of β-catenin gene of Helicoverpa armigera was obtained by RACE method and named as Har-β-catenin (Gen Bank accession number KJ206237). Its open reading frame is 2382 bp, encoding 793 amino acid residues. Similar to other reported β-catenin proteins, the har-β-catenin protein has multiple ARM repeat domains. The expression of Har-β-catenin gene in diapause and non-diapause pupa brains was compared by RT-PCR. The results showed that the expression of Har-β-catenin gene in non-diapause pupa brain was higher than that in the diapause pupae. Finally, we constructed an eukaryotic expression vector containing the full-length coding region of Har-beta-catenin and investigated the subcellular localization of Har-beta-catenin. These results provide a basis for our further study on the role of Wnt / β-catenin signaling in diapause of cotton bollworm.
