目的观察凝血酶对人肝癌细胞株HepG2增殖的影响及其诱导凋亡的作用。方法体外培养人肝癌细胞HepG2,MTT法观测不同浓度凝血酶在不同时间点对肝癌细胞增殖的影响,TUENL法检测其诱导凋亡作用。结果高浓度凝血酶对肝癌细胞增殖有明显抑制作用(P<0.05),当浓度大于100U/ml作用时间在48h后,其抑制率最高可达到(39.08±2.32)%,且不随浓度的提高和时间的延长而增加;高浓度凝血酶能够诱导肝癌细胞凋亡,当浓度大于100U/ml且作用时间大于48h后,各组凋亡指数无显著性差异(P>0.05)。结论高浓度凝血酶能够明显抑制体外生长的人肝癌细胞株HepG2并诱导肝癌细胞凋亡,且存在一定时间(48h内)及浓度(﹤100U/ml)依赖性。
Objective To investigate the effect of thrombin on the proliferation of human hepatocellular carcinoma cell line HepG2 and its role in inducing apoptosis. Methods HepG2 cells were cultured in vitro. MTT assay was used to observe the effects of different concentrations of thrombin on the proliferation of hepatocellular carcinoma cells at different time points. TUENL assay was used to detect the apoptosis of hepatocellular carcinoma cells. Results High concentration of thrombin had a significant inhibitory effect on the proliferation of hepatoma cells (P <0.05). When the concentration was higher than 100 U / ml for 48 h, the highest inhibitory rate was (39.08 ± 2.32)%, (P> 0.05). The apoptotic index of hepatocarcinoma cells was increased with high concentration of thrombin. When the concentration was more than 100U / ml and the action time was longer than 48h, there was no significant difference (P> 0.05). Conclusion High concentration of thrombin can significantly inhibit the growth of human hepatoma HepG2 cells in vitro and induce apoptosis of hepatocellular carcinoma cells in a certain period of time (within 48h) and concentration (<100U / ml).
