为探讨TPO(血小板生成素)基因导入对T细胞增殖和分化影响的规律,将编码人TPO的质粒DNA60μg用脂质体包裹后,经肌肉途径转导入Balb/c小鼠体内。用CD4和CD8的特异性抗体标记小鼠各种免疫组织中的T细胞,借助流式细胞术分析它们的组成和变化。结果发现TPO基因导入使骨髓中的成熟和未成熟T细胞数量在经过短期下降后全部大幅度增多,胸腺未成熟T细胞增加;而脾脏和血液循环中成熟T细胞增多,特别是在血液中CD4阳性细胞的增加最为明显。这些特点大约在基因导入后两周内形成,并持续至少两周以上。本文所得的结果提示TPO是一种免疫促进分子,TPO基因导入后小鼠血液中多种淋巴因子的升高与它过量表达对T细胞的刺激有关。
In order to investigate the regularity of TPO (thrombopoietin) gene transfection on T cell proliferation and differentiation, 60 μg of plasmid DNA encoding human TPO was encapsulated in liposome and then transplanted into Balb / c mice via muscle pathway. T cells in various mouse immune tissues were labeled with specific antibodies to CD4 and CD8, and their composition and changes were analyzed by flow cytometry. The results showed that the introduction of TPO gene in bone marrow so that the number of mature and immature T cells in the short-term decline after all a substantial increase in immature thymus T cells increased; and spleen and blood circulation increased in mature T cells, especially in the blood CD4 The increase of positive cells is the most obvious. These traits are formed approximately two weeks after gene introduction for at least two weeks. The results obtained in this paper suggest that TPO is an immune-promoting molecule. The increase of various lymphokines in the blood of mice after TPO gene is introduced is related to its stimulation of T cell overexpression.
