目的研究常见化疗药物对人肺癌株A549 livin基因的影响。方法将化疗药按不同浓度100、50、25、12.5、6.25、3.13、1.56、0.78μg/ml加入A549后,分别培养24、487、2 h,用MTT比色法检测细胞凋亡,RT-PCR检测livin mRNA的表达,用Western blot法检测livin蛋白的变化。结果长春新碱、多柔比星、卡铂刺激人肺癌株24、48 h后,细胞凋亡率随化疗药物的浓度增加而升高,livin基因的表达明显减少(P<0.01);作用72 h后livin基因表达量开始增加。依托铂苷作用A549,24、487、2 h后,livin基因的表达均随药物浓度的增加逐渐下降。环磷酰胺作用24、48、72 h后livin基因的表达无明显变化(P>0.05)。结论环磷酰胺肺对癌株A549不敏感,长春新碱、多柔比星、卡铂抑制肺癌细胞A549 livin基因表达和细胞增殖,促进细胞凋亡。
Objective To study the effect of common chemotherapeutic drugs on the livin gene of human lung cancer A549 cells. Methods The chemotherapeutic agents were added into A549 cells at various concentrations of 100, 50, 25, 12.5, 6.25, 3.13, 1.56 and 0.78 μg / ml respectively and cultured for 24,487 and 2 h respectively. Cell apoptosis was detected by MTT colorimetric assay. PCR was used to detect the expression of livin mRNA, and the changes of livin protein were detected by Western blot. Results After vincristine, doxorubicin and carboplatin stimulated human lung cancer cells for 24 and 48 h, the apoptosis rate increased with the increase of chemotherapeutics and the expression of livin gene was significantly decreased (P <0.01) After livin gene expression began to increase. After A549, 24, 487 and 2 h treatment with glycoside, the expression of livin gene decreased gradually with the increase of drug concentration. Cyclophosphamide effect 24, 48, 72 h after livin gene expression no significant change (P> 0.05). Conclusion Cyclophosphamide is insensitive to A549 cells. Vincristine, doxorubicin and carboplatin inhibit the livin gene expression and cell proliferation in lung cancer A549 cells and promote cell apoptosis.
