目的:观察白细胞介素-6(IL-6)和血清E2对绝经后骨质疏松症的影响,并探讨IL-6与E2的关系。方法:行开腹输卵管峡部结扎后切除双侧卵巢建立绝经后骨质疏松大鼠模型,双抗夹心酶联免疫吸附法(ELISA)检测血清和骨组织培养液(RPML1640)中IL-6含量,血清E2用RIA法测定,同步用IBAS计算机全自动图象分析系统对不脱钙组织行形态计量学指标测量。结果:去势后早期(≤4周)即有血中和骨组织中IL-6升高和E2下降(P<0.05)。随着去势时间延长(4-22周),骨组织中IL-6浓度高于血中浓度(P<0.05),血中IL-6与E2呈高度负相关(r=-0.63,P<0.01),并出现骨小梁体积减小和骨小梁平均厚度减少以及骨吸收表面增大等骨质疏松形态计量学参数变化(P均<0.01)。去势大鼠血或骨组织中IL—6浓度在术后4—22周之间升高的程度无显著性,而骨组织中IL-6与骨形态计量学各参数的相关程度依次为RS>MTT>FS>TBV。结论:IL-6可能参与骨质疏松症的发生和发展并造成骨组织形态的改变。
Objective: To observe the effects of interleukin-6 (IL-6) and serum E2 on postmenopausal osteoporosis and to explore the relationship between IL-6 and E2. Methods: The ovariectomized rats were established by excision of bilateral ovaries after laparotomy of isthmus. The levels of IL-6 in serum and bone tissue culture fluid (RPML1640) were detected by double-antibody sandwich enzyme-linked immunosorbent assay (ELISA) Serum E2 was measured by RIA method, simultaneously with IBAS computer automatic image analysis system for non-decalcified histomorphometry measurement. Results: In the early period after castration (≤4 weeks), there was an increase of IL-6 and a decrease of E2 in blood and bone (P <0.05). With prolonged castration (4-22 weeks), IL-6 concentration in bone tissue was higher than that in blood (P <0.05), and IL-6 in blood was highly negatively correlated with E2 (r = -0. 63, P <0.01). The morphological parameters of osteoporosis such as decreased trabecular bone volume and average trabecular thickness, and increased bone resorption surface (P <0.01). The level of IL-6 in blood or bone tissue of ovariectomized rats did not increase significantly between 4 and 22 weeks after operation, while the correlation between IL-6 and the parameters of bone histomorphometry was RS > MTT> FS> TBV. Conclusion: IL-6 may be involved in the occurrence and development of osteoporosis and cause changes of bone tissue morphology.
