背景与目的Rho家族蛋白与肿瘤细胞的生长、分化、转移有着密切的关系。本研究通过观察RhoE在肺癌、乳腺癌以及相应癌旁组织中的表达情况及其与临床病理分级的关系,为探索RhoE的生物学功能和临床意义提供依据。方法运用免疫组化SABC法检测62例肺癌和34例乳腺癌及相应癌旁组织中RhoE的表达水平。结果RhoE在乳腺和肺正常组织中普遍表达,但在肿瘤组织表达低下或缺失,而且这种差异在乳腺组织中要高于肺组织。RhoE在乳腺癌组织中的染色平均值为3.65±0.62,在相应癌旁组织中为10.53±0.44,明显高于肿瘤组织(t=12.402,P<0.001);在肺癌组织中的染色平均值为2.19±0.19,在癌旁组织中为4.11±0.24,高于相应肿瘤组织(t=7.123,P<0.005),并且随着肿瘤分化程度的降低染色强度有下降的趋势(x~2=26.536,P<0.005)。结论RhoE在肿瘤组织中表达下降,结合前期实验结果及国外新近的研究成果,可以认为RhoE在肺癌和乳腺癌的发生发展中具有明显的负调控作用,因此对RhoE进行进一步研究将为肿瘤的分子靶向治疗提供新的靶点。
Background and Objective Rho family proteins are closely related to the growth, differentiation and metastasis of tumor cells. In this study, by observing the expression of RhoE in lung cancer, breast cancer and corresponding paracancerous tissues and its relationship with clinicopathological grade, this study provides a basis for exploring the biological function and clinical significance of RhoE. Methods Immunohistochemical SABC method was used to detect the expression of RhoE in 62 cases of lung cancer and 34 cases of breast cancer and corresponding paracancerous tissues. Results RhoE was generally expressed in normal breast and lung tissues but low or absent in tumor tissues, and this difference was higher in breast tissues than in lung tissues. RhoE in breast cancer tissue staining average of 3.65 ± 0.62, corresponding paracancerous tissue was 10.53 ± 0.44, significantly higher than the tumor tissue (t = 12.402, P <0.001); in lung cancer tissue staining average 2.19 ± 0.19, 4.11 ± 0.24 in paracancer tissues, which was higher than that in corresponding tumor tissues (t = 7.123, P <0.005). The staining intensity decreased with the degree of tumor differentiation (x ~ 2 = 26.536, P <0.005). Conclusions The expression of RhoE is decreased in tumor tissue. Combined with the results of previous studies and recent studies abroad, RhoE may be considered as a negative regulator of lung cancer and breast cancer.
