采用不同蛋白酶酶解乳清浓缩蛋白,确定胰蛋白酶是优良的酶源。通过单因素和响应面回归分析,得到酶解工艺的最佳参数为:底物浓度30mg/mL、水解时间90min、温度50℃、加酶量2%(w/w)、pH7.6。在优化的酶解条件下,水解度约为19%,10mg/mL酶解产物的DPPH自由基清除率为78.78%,IC50为5.52mg/mL,显示出较好的抗氧化活性。高效液相排阻色谱(HPSEC)分析表明,酶解产物的相对分子质量分布在300-2600,主要存在的是1000以下的短肽。
Using different proteases to digest whey protein concentrate, trypsin is an excellent enzyme source. Through single factor and response surface regression analysis, the optimal parameters of enzymolysis process were as follows: substrate concentration 30mg / mL, hydrolysis time 90min, temperature 50 ℃, enzyme dosage 2% (w / w), pH7.6. Under optimal conditions, the degree of hydrolysis was about 19%. The DPPH radical scavenging rate of 10 mg / mL hydrolyzate was 78.78% and the IC50 was 5.52 mg / mL, which showed good antioxidant activity. High performance liquid chromatography (HPSEC) analysis showed that the relative molecular weight of the hydrolyzate ranged from 300 to 2600, and the short peptides below 1000 were mainly present.
