采用免疫组化S-P法、Feulgen染色及图像分析技术检测50例原发性乳腺浸润性导管癌雌激素调节Cath-D表达情况,细胞核形态和DNA含量。结果显示,50例乳腺癌中,实质Cath-D高表达率为36%,间质Cath-D高表达率为56% ;Cath-D表达与ER表达比较,无相关性(P>0.05);Cath-D与有无淋巴转移的乳腺癌比较,两者实质高表达率无显著性差异(p>0.05),两者间质Cath-D 高表达率有显著性差异(p<0.05);Cath-D与Ⅰ、Ⅱ、Ⅲ 级乳腺癌比较,两者实质及间质Cath-D高表达率均无显著性差异(p>0.05)。乳腺癌图像分析发现,随着组织学分级的增加,DI值、SPF值(S期细胞比率)、异倍体检出率及细胞形态参数均逐渐增大,均有显著性差异(p<0.05)。上述结果表明,间质Cath-D表达与肿瘤转移相关,其高表达常提示乳腺癌的高转移率。乳腺癌细胞核形态参数和DNA含量可作为乳腺癌诊断、预后判定的指标之一。
Using immunohistochemical S-P method, Feulgen staining, and image analysis techniques, 50 cases of primary breast invasive ductal carcinomas were treated with estrogen to regulate Cath-D expression, nuclear morphology and DNA content. The results showed that in the 50 cases of breast cancer, the high expression rate of Cath-D in the parenchyma was 36%, and the expression of Cath-D in the interstitial cells was 56%. There was no correlation between Cath-D expression and ER expression (P>0.05). There was no significant difference in the high expression rate of Cath-D between breast cancer and lymph node metastasis (p>0.05). There was a significant difference in Cath-D expression between the two groups (p<0.05). There was no significant difference in the expression of Cath-D between the two groups (P>0.05). Analysis of breast cancer images showed that as the histological grade increased, DI value, SPF value (S-phase cell ratio), heteroploid detection rate, and cell morphology parameters all gradually increased, with significant differences (p<0.05). . The above results indicate that Cath-D expression in the interstitium is associated with tumor metastasis, and its high expression often suggests a high metastatic rate in breast cancer. Nuclear morphological parameters and DNA content of breast cancer can be used as indicators of diagnosis and prognosis of breast cancer.
