目的研究 RNA 干扰 Notch1基因对 T 细胞急性淋巴细胞白血病细胞株增殖的影响,并探讨其作用机制。方法根据 Notch1-1C 基因表达序列设计 RNA 干扰片断,筛选有效的片断应用脂质体转染 T-ALL 细胞株 SupT1和Jurkat 细胞。RT-PCR 检测 Notch1和 Hes1基因的表达;MTT 法测定细胞生长曲线;免疫细胞化学测定 Notch1蛋白的表达;流式细胞仪检测细胞周期及其凋亡。结果
Objective To study the effect of Notch1 RNA interference on the proliferation of T cell acute lymphoblastic leukemia cell line and to explore its mechanism. Methods According to the sequence of Notch1-1C gene, RNA interference fragments were designed and valid fragments were screened for liposome transfection of T-ALL cell lines SupT1 and Jurkat cells. The expression of Notch1 and Hes1 was detected by RT-PCR. The cell growth curve was measured by MTT assay. The expression of Notch1 protein was detected by immunocytochemistry. Cell cycle and apoptosis were detected by flow cytometry. result
