[目的]观察孕期十溴联苯醚(Decabromodiphenyl ether,BDE-209)暴露对子代大鼠活动能力及空间学习记忆能力的影响。[方法]将SD孕鼠随机分为对照组和100、300、900mg/kg BDE-209染毒组。于妊娠第6~20天期间,3个染毒组分别每天灌胃BDE-209染毒,对照组采用玉米油灌胃,连续15d。采用旷场实验观察子代大鼠活动能力,采用Morris水迷宫实验检测大鼠空间学习及记忆能力。[结果](1)旷场实验表明:各组子代大鼠在旷场中活动的总距离差异无统计学意义(雌性子代大鼠,F=2.13,P>0.05;雄性子代大鼠,F=0.70,P>0.05)。(2)Morris水迷宫实验表明:在隐蔽平台实验中,训练第3天,雌性子代大鼠的300mg/kg和900mg/kg染毒组与对照组相比,逃避潜伏期延长,差异具有统计学意义(P<0.01);而雄性子代大鼠各组差异无统计学意义(P>0.05)。在空间探索实验中,雄性子代大鼠各组间平台象限活动时间的差异有统计学意义(F=4.17,P<0.01),其中100mg/kg和300mg/kg染毒组与对照组相比,平台象限活动时间缩短,其差异有统计学意义(P<0.01);而雌性子代大鼠各组间差异无统计学意义(P>0.05)。[结论]孕期BDE-209暴露对子代大鼠活动能力无明显影响,可能导致雌性子代大鼠空间学习记忆能力下降和雄性子代大鼠空间位置记忆能力下降。
[Objective] To observe the influence of decabromodiphenyl ether (BDE-209) exposure during pregnancy on activity and spatial learning and memory of offspring rats. [Methods] Pregnant SD rats were randomly divided into control group and 100, 300, 900mg / kg BDE-209 exposure group. During the 6th to 20th days of gestation, the three exposure groups were orally administered with BDE-209, while the control group were fed with corn oil for 15 days. The open-field test was used to observe the activity of offspring rats. Morris water maze test was used to detect spatial learning and memory ability. [Results] (1) Open-field experiments showed that there was no significant difference in the total distance of the offspring rats in each progeny group (female offspring, F = 2.13, P> 0.05) , F = 0.70, P> 0.05). (2) Morris water maze test showed that in the hidden platform experiment, on the 3rd day of training, the escape latency of 300 mg / kg and 900 mg / kg in female offspring rats was longer than that of the control group, the difference was statistically significant (P <0.01), while the male offspring rats in each group showed no significant difference (P> 0.05). In the space exploration experiment, there were significant differences in the platform quadrant activity time among male offspring rats in each group (F = 4.17, P <0.01). Compared with the control group, the 100mg / kg and 300mg / kg exposure groups (P <0.01). There was no significant difference between the female offspring rats in each group (P> 0.05). [Conclusion] There is no significant effect of BDE-209 exposure during pregnancy on the activity of offspring rats, which may lead to the decline of spatial learning and memory ability of female offspring and the decrease of spatial memory of male offspring.
