目的:分析不同生长年限白芍及不同组分白芍多糖中多糖的单糖组成及摩尔比。方法:建立PMP柱前衍生化HPLC法测定不同生长年限的白芍以及采用分级醇沉法得到的白芍多糖不同组分中多糖的单糖组成和摩尔比。结果:白芍多糖中的单糖主要是由甘露糖、鼠李糖、葡萄糖、阿拉伯糖、半乳糖及木糖组成,摩尔比为:156.26∶0.73∶0.85∶1.03∶1.17∶0.79。随着生长年限的增加,白芍多糖中葡萄糖摩尔百分比逐渐增加,其余5种单糖摩尔百分比逐渐降低;白芍多糖不同组分中葡萄糖的含量随着多糖分子量的减小,葡萄糖摩尔百分比逐渐增加,其余5种单糖摩尔百分比逐渐降低。结论:PMP柱前衍生化HPLC法可用于白芍多糖中单糖组成分析,不同生长年限白芍及不同组分白芍多糖中单糖摩尔比不同,葡萄糖是构成白芍多糖的主要单糖。
OBJECTIVE: To analyze the monosaccharide composition and molar ratio of polysaccharides in Radix Paeoniae Alba and Radix Paeoniae Alba by different growth stages. Methods: PMP pre-column derivatization was established for the determination of monosaccharide composition and molar ratio of different growth stages of white peony root and polysaccharide from different fractions of the polysaccharide of Paeonia lactiflora. Results: The monosaccharide in the polysaccharides of Paeonia lactiflora mainly consisted of mannose, rhamnose, glucose, arabinose, galactose and xylose with the molar ratio of 156.26: 0.73: 0.85:1.03: 1.17: 0.79. With the increase of the growth period, the mole percentage of glucose in the polysaccharides of Paeonia lactiflora increased gradually and the mole percentages of the other five kinds of monosaccharides decreased gradually. The content of glucose in different fractions of the polysaccharides of Paeonia lactiflora increased gradually as the molecular weight of the polysaccharides decreased, , The other five kinds of monosaccharide mole percentage gradually decreased. Conclusion: The PMP pre-column derivatization HPLC method can be used for the monosaccharide composition analysis of polysaccharides from Paeonia lactiflora. The molar ratio of monosaccharides in Paeonia lactiflora and Radix Paeoniae Alba is different in different growth periods. Glucose is the main monosaccharide which constitutes the polysaccharide of Paeonia lactiflora.
