近年来,有关合并用药后药物相互作用的报告逐年增加,其中药物的不良反应和有害作用占相当比例,文献中巳出现许多因药物相互作用而引起死亡的例子。因此,在药剂研制和临床应用时,必须注意合并用药中相互作用的问题。药物相互作用的机制非常复杂。大体上可分如下几类:1.在吸收、分布、代谢、排泄过程中的相互作用(下称ADME作用):指一个药物可影响另一个药物的ADME作用,包括影响药物的胃肠道吸收;影响药物与血浆蛋白的结合;激活或抑制药物代谢,增加或减少药物从小便中排出。2.药物在药理方面的相互作用:两种药物可能因相加或拮抗的药理作用而在合并用药时造成不良反应,3.其他:
