目的观察经原核表达得到的小鼠β-防御素-1(mBD-1)对Hek293细胞中CCR6-CCL20通路的影响以及对促进Hek293细胞趋化移动的情况。方法用定量PCR方法检测CCR6和CCL20的基因表达情况,同时用Transwell小室研究其对Hek293细胞的定向移动作用。结果 mBD-1可以使CCR6的表达量增加,且mBD-1浓度越高时CCR6表达量越高,差异有统计学意义(P<0.05)。CCL20的表达量不随mBD-1浓度的升高而变化。同时mBD-1可以促进Hek293细胞作定向移动。结论本研究为进一步研究mBD-1对细胞的免疫诱导机制奠定基础。
Objective To observe the effects of mBD-1 on the CCR6-CCL20 pathway in Hek293 cells and to promote the chemotactic migration of Hek293 cells. Methods The gene expression of CCR6 and CCL20 was detected by quantitative polymerase chain reaction (RT-PCR). The translocation of CCR6 and CCL20 was also studied by Transwell chamber. Results mBD-1 increased the expression of CCR6, and the higher the mBD-1 concentration, the higher the CCR6 expression (P <0.05). The expression of CCL20 did not change with the increase of mBD-1 concentration. At the same time mBD-1 can promote Hek293 cells for directional movement. Conclusion This study lays the foundation for further study on the mechanism of mBD-1 immune induction.
