目的:分析基于胃窦肌间Cajal间质细胞(Myenteric Interstitial Cells of Cajal,ICC-MY)自噬研究伴抑郁的功能性消化不良(Functional Dyspepsia,FD)的发病机制。方法:20只SD大鼠随机分为正常对照组、模型组,每组10只。模型组采用慢性夹尾激惹刺激法制备FD大鼠模型。透射电镜观察两组大鼠胃窦肌间Cajal间质细胞内自噬体数量;免疫荧光法检测两组ICC-MY内Beclin1、微管相关蛋白1轻链3 B(microtubule-associated protein 1 light chain 3B,LC3B)蛋白的荧光强度。结果:与正常对照组比较,模型组大鼠胃窦ICC-MY内自噬体数量多,Beclin1、LC3B蛋白的荧光强度显著升高(均P=0.000)。结论:伴抑郁的FD的发病机制可能与抑郁引起胃窦ICC-MY过度自噬有关。
OBJECTIVE: To investigate the pathogenesis of functional dyspepsia (FD) based on autophagy based on Myenteric Interstitial Cells of Cajal (ICC-MY). Methods: Twenty SD rats were randomly divided into normal control group and model group with 10 rats in each group. The model group was treated with chronic noxious stimulation to prepare FD rat model. The numbers of autophagosomes in Cajal interstitial cells in the antrum of the antrum of the two groups were observed by transmission electron microscopy. The expressions of Beclin1, microtubule-associated protein 1 light chain 3B, LC3B) protein. Results: Compared with the normal control group, the number of autophagosomes in the ICC-MY rat model of gastric antrum was increased in the model group, and the fluorescence intensity of Beclin1 and LC3B proteins was significantly increased (both P = 0.000). Conclusion: The pathogenesis of FD with depression may be related to the over-autophagy of ICC-MY in gastric antrum induced by depression.
