目的 了解基质降解对原发性肝癌的作用。方法 33例原发性肝细胞癌患者术中取癌组织及癌旁组织 ;提取组织总RNA ;用逆转录共扩增定量PCR方法测定明胶酶A (MMP2 )基因表达水平。结果 肝癌组织明胶酶A基因表达水平 (3 2 0± 0 34 )明显高于癌旁的肝硬化组织 (0 70±0 16 )及正常肝组织 (0 34± 0 0 7)。肝癌患者MMP2 基因表达水平与血清AFP浓度、肿瘤大小无关。结论 明胶酶A在原发性肝癌的进展中可能起重要作用。
Objective To understand the effect of matrix degradation on primary liver cancer. Methods Totally 33 cases of primary hepatocellular carcinoma (HCC) were treated with cancer tissue and para-cancerous tissues. Total RNA was extracted from tissue samples. Gelatinase A (MMP2) gene expression was determined by reverse transcription-polymerase chain reaction (RT-PCR) Results The level of gelatinase A gene expression in hepatocellular carcinoma was significantly higher than that in adjacent cirrhosis (0 70 ± 0 16) and normal liver tissue (0 34 ± 0 0 7) (3 2 0 ± 0 34). The expression level of MMP2 gene in patients with hepatocellular carcinoma has nothing to do with serum AFP concentration and tumor size. Conclusion Gelatinase A may play an important role in the progression of primary liver cancer.
