目的利用免疫磁珠法分选并鉴定人前列腺癌类干细胞。方法利用免疫磁珠法从人前列腺癌细胞株PC3中分选CD133+/CD44+细胞,通过免疫组化、流式细胞术、CCK8、平板克隆形成实验及裸鼠成瘤实验鉴定其表面标志物表达情况及肿瘤干细胞特性。结果经免疫磁珠法分选所得CD133+/CD44+细胞比例为0.50%;免疫组化及流式显示其CD44/CD133/整合素α2β1均高表达;其细胞增殖和克隆形成率高于PC3细胞;以低密度注射时,其裸鼠体内成瘤率明显高于PC3细胞。结论利用免疫磁珠法可从人前列腺癌细胞株PC3中高效的分选出具有肿瘤干细胞生物学特性的前列腺癌类干细胞。
Objective To isolate and identify human prostate cancer stem cells by immunomagnetic beads method. Methods CD133 + / CD44 + cells were sorted from human prostate cancer cell line PC3 by immunomagnetic beads method and their surface markers were identified by immunohistochemistry, flow cytometry, CCK8, plate clone formation assay and tumorigenesis in nude mice And tumor stem cell characteristics. Results The percentage of CD133 + / CD44 + cells sorted by immunomagnetic beads method was 0.50%. The expression of CD44 / CD133 / integrin α2β1 was highly expressed by immunohistochemistry and flow cytometry. The rate of cell proliferation and colony formation was higher than that of PC3 cells. Low density injection, the nude mice tumor formation rate was significantly higher than PC3 cells. Conclusion Immunomagnetic beads method can be used to efficiently isolate prostate cancer stem cells with tumor stem cell biological characteristics from human prostate cancer cell line PC3.
