共价闭合环状脱氧核糖核酸(cccDNA)是嗜肝DNA病毒基因组的复制中间体,也是嗜肝病毒持续感染的关键因素,它的长期存在也是乙型肝炎慢性化的主要原因之一。随着分子生物学技术的发展,HBV cccDNA的检测方法也在不断改进,准确地检测肝内和/或血清HBV cccDNA的量对病情判断、疗效评估和预后均有重要意义。本文对cccDNA的形成过程、临床检测方法、意义及存在的问题进行了综述。
Covalent closed circular deoxyribonucleic acid (cccDNA) is a replication intermediate of hepadnavirus genome and is also a key factor for persistent infection with hepadnavirus. Its long-term presence is also one of the major causes of chronic hepatitis B infection. With the development of molecular biology technology, the detection methods of HBV cccDNA are also constantly improving. The accurate detection of the amount of HBV cccDNA in the liver and / or serum is of great significance for disease judgment, curative effect evaluation and prognosis. In this paper, the formation process of cccDNA, clinical testing methods, significance and existing problems are reviewed.
