目的探讨中药骨碎补对体外培养的人牙龈成纤维(HGF)细胞超微结构、碱性磷酸酶(ALP)活性的影响。方法2005年9月至11月于河北医科大学第四医院实验中心将体外培养的HGF细胞分别与质量浓度为10、50、100、500、1000μg/mL的骨碎补共同孵育,四甲基偶氮唑盐(MTT)比色法测定ALP活性,选出ALP活性最强的骨碎补质量浓度作为最佳浓度,并作用于培养细胞。电镜下观察细胞的超微结构改变。结果在骨碎补各质量浓度范围内,实验组ALP活性均高于对照组(P均<0.05),骨碎补质量浓度为100μg/mL时ALP活性最强。与对照组比较,实验组细胞器数量增加,细胞体较大,有丰富的胞浆,胞浆内粗面内质网数量明显增多,线粒体较为发达,细胞核及核仁体积较大。结论骨碎补在一定质量浓度范围内可使ALP活性增强,细胞器数量增加,功能增强。
Objective To investigate the effect of Chinese herbal medicine, Rhizoma Drynariae, on the ultrastructure and alkaline phosphatase (ALP) activity of human gingival fibroblast (HGF) cells cultured in vitro. METHODS: From September to November 2005, the experimental HGF cells from the Fourth Hospital of Hebei Medical University were incubated with in vitro cultured HGF cells with 10, 50, 100, 500, and 1000 μg/mL concentrations of rhododendron. The ALP activity was determined by MTT colorimetric method, and the mass concentration of A. crunifolia with the highest ALP activity was selected as the optimal concentration, and it was applied to cultured cells. The ultrastructural changes of the cells were observed under electron microscope. Results The ALP activity in the experimental group was higher than that in the control group (P<0.05), and the ALP activity was the strongest at the mass concentration of 100 μg/mL. Compared with the control group, the number of organelles in the experimental group increased, the cell body was large, and there was abundant cytoplasm. The number of rough endoplasmic reticulum in the cytoplasm increased significantly, the mitochondria were more developed, and the nucleus and nucleoli were relatively large. Conclusion In the certain mass concentration range, Rhizoma Drynariae can increase the activity of ALP, increase the number of organelles and enhance the function.
